搖床
搖(yao)(yao)床又稱搖(yao)(yao)瓶(ping)機,它是培(pei)養好氣性微生物的小(xiao)型(xing)試驗設備或(huo)作為種(zhong)子擴(kuo)大(da)培(pei)養之用,常用的搖(yao)(yao)床有(you)往復(fu)式和旋(xuan)轉(zhuan)式兩種(zhong)。往復(fu)式搖(yao)(yao)床的往復(fu)頻(pin)率(lv)一般(ban)在(zai)80~140次/min,沖程一般(ban)為5~14cm,如頻(pin)率(lv)過快(kuai)、沖程過大(da)或(huo)瓶(ping)內液體裝量過多,在(zai)搖(yao)(yao)動時液體會濺到(dao)包扎瓶(ping)口的紗布(bu)或(huo)棉塞上,導致雜菌污染(ran),特(te)別是啟動時更容(rong)易發生這(zhe)種(zhong)情(qing)況。旋(xuan)轉(zhuan)式搖(yao)(yao)床的偏心距一般(ban)在(zai)3~6cm之間(jian),旋(xuan)轉(zhuan)次數為60~300rpm。
放在搖(yao)床上的(de)(de)培養瓶(ping)(一般(ban)為三(san)角瓶(ping))中的(de)(de)發酵(jiao)液所(suo)需要(yao)的(de)(de)氧是由(you)空氣(qi)經(jing)瓶(ping)口(kou)(kou)包扎的(de)(de)紗布(bu)(一般(ban)8層)或棉(mian)塞通入的(de)(de),所(suo)以氧的(de)(de)傳遞與瓶(ping)口(kou)(kou)的(de)(de)大小、瓶(ping)口(kou)(kou)的(de)(de)幾何形狀、棉(mian)塞或紗布(bu)的(de)(de)厚度和密度有關(guan)。在通常情況下,搖(yao)瓶(ping)的(de)(de)氧吸收系(xi)數取決于搖(yao)床的(de)(de)特性和三(san)角瓶(ping)的(de)(de)裝(zhuang)樣(yang)量。

液體(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)接(jie)(jie)種(zhong)(zhong)(zhong)箱是(shi)專為移接(jie)(jie)液體(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)而設(she)(she)計的。比(bi)固體(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)箱窄長,單側兩人操作。內(nei)設(she)(she)軌道和紫外(wai)線燈,箱兩端開有(you)(you)(you)高25cm,寬(kuan)10cm的長方形(xing)出口(kou),方便(bian)菌(jun)(jun)種(zhong)(zhong)(zhong)進(jin)(jin)出,洞口(kou)設(she)(she)有(you)(you)(you)小(xiao)推(tui)門。進(jin)(jin)出口(kou)下處設(she)(she)蒸汽(qi)源,接(jie)(jie)種(zhong)(zhong)(zhong)時用蒸汽(qi)封(feng)住進(jin)(jin)出口(kou),以(yi)防雜菌(jun)(jun)進(jin)(jin)入箱內(nei)。箱背面設(she)(she)有(you)(you)(you)液體(ti)菌(jun)(jun)種(zhong)(zhong)(zhong)移接(jie)(jie)管(guan)能進(jin)(jin)入的小(xiao)孔。
接(jie)(jie)種箱滅菌(jun)(jun)時,用(yong)紫外(wai)線照射30min。如(ru)果(guo)沒有(you)紫外(wai)線燈(deng),可(ke)用(yong)甲醛(quan)(quan)和高(gao)(gao)錳酸鉀(甲醛(quan)(quan)10~14mL/m3+高(gao)(gao)錳酸鉀5~7g/m3空(kong)間)熏蒸30min以上。使用(yong)時,先將所需(xu)物(wu)品和工具放(fang)入接(jie)(jie)種箱內(nei),然后進(jin)行(xing)藥劑熏蒸和紫外(wai)線滅菌(jun)(jun),再按無菌(jun)(jun)操(cao)作(zuo)進(jin)行(xing)接(jie)(jie)種。接(jie)(jie)種箱的(de)結構(gou)簡單,造價低廉,易消(xiao)毒滅菌(jun)(jun),操(cao)作(zuo)方便,而且人(ren)在箱外(wai)操(cao)作(zuo),氣溫較高(gao)(gao)時也能作(zuo)業(ye)。缺點是進(jin)出培養基費工費時,每(mei)次接(jie)(jie)種前都需(xu)要進(jin)行(xing)滅菌(jun)(jun)。
電冰柜 微生物實驗室的冰箱主要有兩種:普通冰箱和低溫冷凍冰箱。普通冰箱一般都具有兩個柜子,即鮮藏柜和冷藏柜,溫度分別為4℃和-20℃;低溫冷凍冰箱溫度一般控制在-40~-80℃。它們都可以用于微生物菌種保藏。鮮藏柜常用于保存斜面菌種,保藏時間在3個月左右。超過3個月,斜面就會變干,因此需要轉接菌種。如果要長時間保存菌種,則需要經過處理后,貯藏于普通冰箱的冷藏柜或低溫冷凍冰箱中,它們的保藏時間較長,一般都在1年以上。
